Emergency crews were called to the corner of Sneydes Road and the Princes Freeway at Werribee about 7.10am after a car hit a light pole.
9News understands one child was sitting on an adult's knee unrestrained, at the time of the accident.
A probationary driver was travelling outbound and smashed into the pole at the centre of the median strip.
Two children have been taken to The Royal Children's Hospital, as well as three people to The Royal Melbourne Hospital.
The accident sparked gridlock traffic, with the closure of all outbound lanes on the Princes Freeway, near Sneydes Road.
Half a dozen traffic lights were also reportedly out throughout Werribee, including the Princes Freeway, due to a power outage in the area.
A silver Mazda SUV has been towed from the scene.
